Bexotegrast is a potent inhibitor of ανβ6 integrin. Bexotegrast can be used for researching fibrosis such as idiopathic pulmonary fibrosis (IPF) and nonspecific interstitial pneumonia (NSIP) (extracted from patent WO2020210404A1, compound 5).

体外研究

Bexotegrass (PLN-74809; 1.82 µM; incubated for 7 days) significantly reduced the expression of type I collagen alpha I (COL1A1) mRNA by 54% in precision cut lung sections (PCLS). Bexotegrass showed a decrease of approximately 50% in Smad2 phosphorylation.

Bexotegrass (1.82 µM; incubation for 3 days) dose dependently reduced Col1a1 mRNA expression in PCLS prepared from fibrotic mouse lungs by up to 71%.

Bexotegrass completely inhibits the adhesion of normal human bronchial epithelial cells to LAP mediated by α v β 6 integrin, with an IC₅₀ of 39.3 nM.

体内研究

Bexotegrass (PLN-74809; oral; 100、250、500 mg/kg； Twice a day; Dose dependent reduction of interstitial fibrocollagen deposition in mice challenged with bleomycin from day 7 to day 21. Bexotegrass dose dependently blocks Smad3 phosphorylation.

Animal Model:	C57BL/6 mice
Dosage:	100, 250, and 500 mg/kg
Administration:	Orally; twice daily; from Day 7 to Day 21
Result:	Showed a dose-dependent, significant reduction in interstitial fibrillar collagen deposition in Bleomycin (3 units/kg)-challenged mice. Dose-dependently blocked Smad3 phosphorylation.
