N-acetyl-D-Leucine is a derivative of D-leucine and a substrate for various enzymes in the amidohydrolase superfamily. It has been used to characterize the function of amidohydrolase enzymes from B. bronchiseptica, S. coelicolor, and G. oxydans.
N-acetyl-D-Leucine is an amino-protecting group N-substituted chiral amino acid. N-acetyl-D-Leucine is a PepT1 and MCT1 inhibitor with IC50 of 0.74 and 11 mM, respectively. N-acetyl-D-Leucine can be used for LysoTracker signaling studies.
体外研究
N-acetyl-D-Leucine is not a substrate of LAT1 and PepT1, but rather an inhibitor of PepT1 with an IC50 of 0.74 mM.
N-acetyl-D-Leucine is a substrate of MCT1 with a Km of 1.0 mM and also an inhibitor of MCT1, with an IC50 of 11 mM.
N-acetyl-D-Leucine (1-5 mM; 7 days) did not show a significant dose-dependent effect on LysoTracker signals in NPC1-deficient Chinese hamster ovary cells, but a statistically significant effect was observed at higher concentrations (5 mM).
体内研究
N-acetyl-D-Leucine (24 mg per rat; intravenous injection; once daily) had no significant effect on the compensation and postural imbalance scores of nystagmus in rats.
