Octamer-binding transcription factor 4 (Oct4) is a transcription factor which, with Sox2, Klf4, and c-Myc, is involved in the reprogramming of somatic cells to produce pluripotent stem cells. OAC3 is an Oct4-activating compound which activates expression through the Oct4 gene promoter. In cells expressing Oct4 with Sox2, Klf4, and c-Myc, OAC3 (1 μM) enhances reprogramming efficiency by increasing the rate of production of induced pluripotent stem cells from embryonic fibroblasts.
OAC3 is a potent Oct4 activator. OAC3 activates Oct4 and Nanog promoters and enhances induced pluripotent stem cells (iPSC) formation. OAC3 facilitates the reprogramming of cells by enhancing efficiency and shortening the reprogramming time.
体外研究
OAC3 (1 μM) enhances reprogramming efficiency by activating Oct4-luc reporter and Nanog-luc reporter in embryonic stem cells (ESCs). OAC3 increases induced pluripotent stem cells (iPSC) generation and accelerates the appearance of iPSC-like colonies. OAC3 (1 μM; 4F (OSKM) infected mouse embryonic fibroblast (MEF)) enhances reprogramming efficiency in iSF1 media.
