N-3-oxo-dodecanoyl-L-Homoserine lactone (3-oxo-C12-HSL) is a bacterial quorum-sensing signaling molecule produced by P. aeruginosa and strains of the B. cepacia complex. It induces the production of IL-8 in 16HBE human bronchial epithelial cells when used at a concentration of 100 µM. Supernatants from 3-oxo-C12-HSL-stimulated 16HBE cells induce chemotaxis of isolated human neutrophils. 3-oxo-C12-HSL has been found in respiratory secretions from patients with cystic fibrosis infected with P. aeruginosa.

体外研究

N-3-oxo-dodecanyl-L-homoserine lactone (25-100 μM, 18 h) inhibits IL-8 expression and induces neutrophil chemotaxis in L828 cells by activating NF - κ B and ERK1/2.
N-3-oxo-dodecanyl-L-homoserine lactone (50 μM, 24 h) induces ER stress response, ROS generation, and mitochondrial membrane potential reduction, leading to caspase 3/8/9/12 activation and inducing apoptosis in BMDM.

N-3-oxo-dodecanoyl-L-homoserine lactone (0.3-1 mg/kg, ip, single dose) enhanced the immune preference of the host, showed immunomodulatory activity in OVA immunized mice model, and improved type 1 diabetes by inhibiting the activation and differentiation of initial T cells.

    以 1 mL 工作液为例,取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中,混合均匀;向上述体系中加入50 μL Tween-80,混合均匀;然后继续加入 450 μL生理盐水定容至 1 mL。
