(S)-4'-nitro-Blebbistatin is a more stable and less phototoxic form of (–)-blebbistatin , which is a selective cell-permeable inhibitor of non-muscle myosin II ATPases. (–)-Blebbistatin rapidly and reversibly inhibits Mg-ATPase activity and in vitro motility of non-muscle myosin IIA and IIB for several species (ICs = 0.5-5 µM), while poorly inhibiting smooth muscle myosin (IC = 80 µM). Through these effects, it blocks apoptosis-related bleb formation, directed cell migration, and cytokinesis in vertebrate cells. However, prolonged exposure to blue light (450-490 nm) results in degradation of blebbistatin to an inactive product via cytotoxic intermediates, which may be problematic for its use in fluorescent live cell imaging applications. The addition of a 4’-nitro group decreases the inherent fluorescence, stabilizes the molecule to circumvent its degradation by prolonged blue light exposure, and decreases its phototoxicity while retaining the in vitro and in vivo activity of (–)-blebbistatin. (S)-4'-nitro-Blebbistatin has the same stereochemistry as the active (–)-blebbistatin enantiomer.

(S)-4'-nitro-Blebbistatin is a derivative of Blebbistatin and an inhibitor of myosin II. (S)-4'-nitro-Blebbistatin is photostable, non-cytotoxic, and non-phototoxic. (S)-4'-nitro-Blebbistatin can serve as an ideal substitute for Blebbistatin to study the role of myosin II in physiology, development, and cell biology[1].

体外研究

(S) -4 '- nitro-Blebistatin (0.31-10 μM; 24 h) can inhibit cytoplasmic division of COS-7 cells, resulting in a multinucleated phenotype with an EC50 of 2.9 μM [1].

(S) -4 '- nitro-Blebistatin (10 μM; 480 nM) was irradiated for 15 minutes; 18 hours) showed no cytotoxicity in HeLa cells, and there was no difference in cell mortality and morphology compared to the control group.

体内研究

(S) -4 '- nitro-Blebistatin (5-20 μM; single dose; 24 hours) In transgenic cldnb: gfp zebrafish embryos, cell migration of the posterior line primordia can be inhibited, resulting in a curved body shape of zebrafish embryos. This effect is similar to Blebbistatin, but the mortality rate of animals in the (S) -4 '- nitro-Blebistatin group is significantly reduced.
