体外研究

Anti-IL11 Antibody (X203) (20 μM; 48 h) can effectively inhibit downstream STAT3 phosphorylation induced by IL-11 in Bleomycin induced mouse nucleus pulposus cells.

Cell Line: Mouse nucleus pulposus (NP) cells treated with Bleomycin (40 μg/mL)

体内研究

Anti-IL11 Antibody (X203) (20 mg/kg, intraperitoneal injection, twice a week starting from 6 weeks of age, lasting for 2.5 weeks) can improve renal function and prolong the lifespan of Alport syndrome (AS) mouse models.

 

Animal Model: Wild-type mice injected with BLM
Dosage: 20 mg/kg
Administration: i.p., alternate days from day 7 to 21 after BLM
Result: Showed no effect on overall survival.
Greatly reduced lung hydroxyproline content.
Reduced the expression of ECM-related genes.
Increased Mmp2/Timp1 ratio (a signature of preferential ECM remodeling) compared to control.
Markedly reduced ERK and SMAD activation whereas STAT3 phosphorylation remained elevated.
