生物活性

Paritaprevir (ABT-450) is a potent non-structural protein 3/4A (NS3/4A) protease inhibitor with EC₅₀s of 1 and 0.21 nM against HCV 1a and 1b, respectively. Paritaprevir is also a SARS-CoV 3CL inhibitor with an IC₅₀ of 1.31 μM.

Paritaprevir demonstrates in vitro antiviral activity against HCV GT1-4 and GT6 (EC₅₀ range, 0.09 to 19 nM), with an EC₅₀ of 0.09 nM against GT4a.

The combination of paritaprevir, ritonavir, ombitasvir (an NS5A protein inhibitor), and dasabuvir (an NS5B non-nucleoside polymerase inhibitor) with or without RBV has been approved to treat HCV genotype 1 infections1.

Solubility: ≥ 2.08 mg/mL (2.72 mM); Clear solution

此方案可获得 ≥ 2.08 mg/mL (2.72 mM，饱和度未知) 的澄清溶液。
以 1 mL 工作液为例，取 100 μL 20.8 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；向上述体系中加入50 μL Tween-80，混合均匀；然后继续加入 450 μL生理盐水定容至 1 mL。
