NIAD-4 is a fluorescent probe that crosses the blood-brain barrier to bind with high affinity to amyloid-β (Aβ) plaques (K = 10 nM). It displays excitation/emission spectra of 475/625 nm, respectively, with the far-red emission occurring upon binding to Aβ.

NIAD-4 can bind to amyloid protein aggregates, hindering their internal rotation and increasing fluorescence yield and intensity, making it an effective tool for detecting amyloid protein. NIAD-4 can cross the brain barrier and, due to its neutral charge and low molecular weight, can be used for imaging amyloid fibrils in the brain. The excitation wavelength of NIAD-4 is 475 nM, and the emission wavelength is 610 nM.
